Cryopreservation of germinal stem cells in rainbow trout: a strategy to entirely restore a genotype of interest

Abstract

Cryobanking of fish genetic resources relies exclusively on sperm cryopreservation, as the cryopreservation of oocytes and embryos is not possible in these species with telolecithal eggs. However, the cryopreservation of germinal stem cells is achievable, because they are structurally less complex than fish oocytes and embryos. In addition, it is shown that when germinal stem cells are injected into the peritoneal cavity of recipient fries, they colonize the fry gonads and undergo gametogenesis, thereby leading to the production of functional gametes. In the course of building the infrastructure of BRC Anim, the animal component of the AgroBRC/RARe infrastructure, a method for germinal stem cell cryopreservation was set up for rainbow trout. Here, we present and comment the final procedure that was adopted: the extenders were devoid of animal products, thawed cell quality was validated, and their ability to colonize recipient gonads was demonstrated. The cryopreservation strategy described enables the cryobanking of biological materials that support the regeneration of a complete genotype of interest after cryopreservation in fish.
